Six Mile is a town in Pickens County, South Carolina, United States. The population was 553 at the 2000 census. It is part of the Greenville–Mauldin–Easley Metropolitan Statistical Area.

According to the United States Census Bureau, the town has a total area of 1.8 square miles (4.7 km²), of which, 1.8 square miles (4.7 km²) of it is land and 0.55% is water.
